Source-derived summary

Lorlatinib, sold under the brand name Lorbrena in the United States, Canada, and Japan, and Lorviqua in the European Union, is an anti-cancer medication used for the treatment of non-small cell lung cancer. It is an orally administered inhibitor of anaplastic lymphoma kinase (ALK) and C-ros oncogene 1 (ROS1), two enzymes that play a role in the development of cancer. It was developed by Pfizer.

The most common adverse reactions include edema, peripheral neuropathy, weight gain, cognitive effects, fatigue, dyspnea, arthralgia, diarrhea, mood effects, hypercholesterolemia, hypertriglyceridemia, and cough.

Lorlatinib was approved for medical use in the United States in November 2018, and in the European Union in May 2019.

Medical uses

Lorlatinib is indicated for the treatment of adults with metastatic non-small cell lung cancer whose tumors are anaplastic lymphoma kinase (ALK)-positive.

Contraindications

Lorlatinib must not be combined with strong inducers (i.e. activators) of the liver enzymes CYP3A4/5 if it can be avoided, as serious cases of liver toxicity have been observed under combination with the CYP3A4/5 inducer rifampicin.

Side effects

The most common side effects in studies were high blood cholesterol (84% of patients), high blood triglycerides (67%), edema (55%), peripheral neuropathy (48%), cognitive effects (29%), fatigue (28%), weight gain (26%), and mood effects (23%). Serious side effects led to dose reduction in 23% of patients and to termination of lorlatinib treatment in 3% of patients.

Editorial summary

This brief starts where responsible research should: with the source description of “Lorlatinib” as chemical compound. Everything that follows is an evidence route, not borrowed authority.

Editorial reviewA sound reference starting point where classification, measurement and the date of the underlying evidence remain visible. The current lead gives the account dated anchors—2018, 2019—that can be checked directly. The selected authority fields contribute no independent date. The account is most persuasive where Lorlatinib, chemical and compound can be independently traced.
Editorial analysis

Why this record matters

The subject matters to the science & nature register because the source frames it as chemical compound. Its deeper value depends on whether names, dates, institutions and citations support that framing.

Evidence profile

Stable identifiers, scientific names and standards terminology offer the best bridge between this overview and specialist evidence. The source revision retrieved here is dated Dec 24, 2025. The linked authority identifier is Q27285820. None of the 0 selected statements returned an explicit reference. The first chronological checks are 2018 and 2019.

Critical limits

Current terminology should not be projected backward without checking the classification used when the underlying evidence was created. The source lead contains qualifying language; that uncertainty should survive quotation, summary and reuse. Authority statements aid reconciliation but still require their own references, qualifiers and ranks to be checked.

How to read it

Check terminology, classification and the date of the cited evidence. Scientific names and technical consensus can change while older records retain historical value.

Three-step research path

  1. Establish the record: confirm the title “Lorlatinib”, its source revision and the description used here.
  2. Expand the search: follow Lorlatinib primary sources, Lorlatinib archive and Lorlatinib research across catalogues and specialist indexes.
  3. Test the account: compare the strongest cited source with the responsible institution’s current record and note any disagreement.

Questions for further research

  1. Which source most directly establishes the central claim about “Lorlatinib”?
  2. Which observation, specimen, dataset or publication supports the account?
  3. Is the terminology current, historical or disputed?
